The clash sees the west Londoners host the Saints for the second time this month after Chelsea battled to a 3-1 Premier League victory over their rivals before the international break.
Chelsea needed a penalty shootout to defeat Aston Villa in the third round last month but will be looking for an easier passage to the fifth round against the Saints.
